Real answer if you're too lazy:

Generally, you go to the con's website, and submit your video as per their rules (regarding format, sending in your form, etc).

Then, they weed through the entries in whatever way they see fit. If you don't get in, then you usually get a nice consolation email and a reminder that your video will be shown in the overflow screening.

If you do get in, then you get an even nicer email saying that you made it.

At the convention itself, the contest varies con-by-con. Some conventions have audience voting, in which they all recieve ballots with their registration info. Other cons have judged voting, or creator voting, or a balance between the two.

Some cons (Anime Boston, for one) offer front-row seating at the competition screening for creators who have made finalist videos, and at most cons the AMV creators usually get together to do stuff.

Then, after the screening is over and the votes are collected, they get tallied. Winners are usually announced during intermission at the Masquerade, and get to go onstage and accept a trophy of some sort, along with seeing their video shown again.
