Parody is clearly protected under fair use, and this has been upheld time and again by the Supreme Court. DMCA takedown notices are filed under penalty of perjury... I'd countersue the Sakura-con and threaten to get their overly zealous lawyer disbarred.

While I can understand wishing to control intellectual property I cannot condone take down actions of parodies if your sole operating premise is roughly "we didn't like what they said"
The later example, which the parody is essentially doing to a degree, could be considered slander/libel (or fraud) which is against the law. I'm not a lawyer though so don't take my word on that.

My example comment had less to do with what is derogatory comments and more to do about making an example of will they pursue every nutter out there who has a bad opinion about Sakura con. I gave no qualifications and made no claims. That statement was complete opinion. The only difference with the youtube parody is that a lot of people were watching it and laughing/talking about it.
The parody is definitely not claiming to be anyone of authority with Sakura con so you are completely wrong there. It could be argued it appears sanctioned by Sakura con if there is no disclaimer but considering it had "The cast of YGOTAS wish they were as Japanese as the guys in this commercial: http://www.youtube.com/watch?v=XP5lz2CYNR4 " in its description and the user is a well known account for posting parodies I don't see an easy way to legally argue that. All of this has nothing to do with the methods they used to get the video down in the first place. Instead of contacting the creator directly they just put a blanket DMCA style thing down which is definitely argueable.
As for defamation there is no way it would be prosecuted. The video made no claims. I don't see anything beyond the catchphrases as opinion/parody which of course could be proven accurate with examples from con that we all know are there =p To me that's the biggest joke here. The video was taken down when it was just as accurate as the original video in describing some anime material or con activities that can be seen at conventions.

I don't think the Parody can do more damage than the commercial did to the Anime Community. It casts all Otaku in a bad light.

At first, I am surprised Dinger hasn't commented, but the more I think about it, it makes sense that he hasn't. You are not about to hear from the AMV staff, or any staffers who disagree with what Sakura-con has done, because these people are already under pressure from within the organization to conform or be put out.
And the emphasis of remix culture at Sakura-con is declining, rapidly. Sakura con has screwed over with timing and room space both AMVs and Cosplay. Normally enemies, the two groups have now formed a rather united front of hatred towards Sakura-con. It seems that that the executive board of Sakura is going a lot more trade showish, a lot like Expo, only with none of its advantages.
I considered going a few months ago, which would have been a rather large chunk of change considering, but what I've kept up with tells me they don't deserve that level of support from me.

That all depends on who is viewing it and where it's being viewed. afaik, if the video is embedded in another website, you can't read the description. I also would have no idea that that particular user posts parodies, and i didn't until you just mentioned it. I also rarely read the description. As i mentioned in my example, you could have little timmy watching it with mommy and daddy who wouldn't know better. You can't say everyone knows it's a parody and Sakura-con had nothing to do with it. There are going to be people out there who don't know better. Honestly, i really don't know what the law is on something like this. I can see how it might be considered defamation or libel or whatever, but the law is so fucked up anyway, who knows. anyway, that doesn't matter... I still disagree with the way it was handled. I just wanted to give a different perspective as to what their motives and justifications were for the take down.
