Making one right now - a yaoi called _Uke On Top_! ^_^ Script is mostly done (estimated length about an hour, rating NC-17), main character designs are done, now moving on to storyboarding, secondary characters, and background art. I was under the impression that the first "anime" or feature was Steamboat Willy done by Walt Disney.
And IIRC Walt is also the person who pioneered alot of the techniques used in anime.
I have heard and seen in print that he is revered as the father of animated films in Japan.
But since the majority of anime is still hand drawn then I would have to say hand drawn.
I would consider anime the film genre and hand drawn the technique to produce anime.

Anime = Japanese Animation to us, but to the Japanese, Anime is any type of animation.
Batman Beyond could be considered an American Anime.

As has been said before, anime in it's true definition is animation, period. So, any form of American animation, whether it's Disney or ABC or Fox, is anime by the dictionary definition.
If you use the subjective definition of anime as Japanese animation, then an american anime is impossible.
Oh, and let's not forget Animatrix, which is now out on DVD and is on my hit list of DVDs to buy.
