Noises Off! for overall funniness - though it definitely helps to have been in some way involved in theatre, I think that made it several times funnier for me.

But the hardest I think I've ever laughed at a movie was the champagne cork-owl scene from Dumb and Dumber... my sides hurt for hours after that.
