For rotoscoping, you are much better off with the original source, meaning a BluRay copy. DVD and TV rips have frame-rates converted, thus blurring occurs. Unless strictly intended, Anime won't have blurring unless they're using life-like camera effects like focus or there's incredibly high motion.

if you are planning to get after effects don't get vegas.... get premiere pro instead. its much better and it works great with after effects. don't listen to the people that say vegas and after effects are better. Agreed. Adobe products shouldn't really be mixed with other softwares. It just distur...

Mac's don't have any decent interfaces for x264. I highly suggest moving the file to a Windows machine with MeGUI installed.
I have a x264 profile sheet I designed which kicks the crap out of pretty much every other method. Lossless video at tiny sizes.
