Yes, we generally do prefer if people read the manual. However, I'll give an explanation.
If you have a version of Premiere that earlier than 6.5, you won't be able to see your effects in the Monitor window because Premiere can't render the effects on the fly. In order to see your effect, just press Enter and you should be prompted to save your file and some other stuff, but after Premiere goes through your timeline and renders everything, you should be able to see the effect. This process is called a "preview."
Premiere 6.5 can do realtime previews where you don't have to press enter and save a little file. It's pretty neat.
